The Technique Of Cellulose Acetate Electrophoresis

The demonstrated method conducted was cellulose acetate electrophoresis, which creates bands to represent contradictory conformation of haemoglobin condition. The width if the bands reveal the amount of hemoglobin that it currently situated in the known sample. As displayed on the diagram there are four Hb variants, which consist of HbC, HbS, HbF, and HbA. Lane four represents the control, and is used to compare the bands from lane one, two and three samples. Lane one has one band in comparison to the other two lanes. In comparison to the control sample, the band resembles for HbA. The thickness of the band also resembles the HbA band in lane 2 however the diagnosis is normal adult of 96-99% concentration. There are two band positions on lane two that have similar traits as HbS and HbA from the control sample. Comparing the two bands in lane two, HbA bands intensity is thicker than the HbS band that is situated above in lane two. The concentration for HbA variant is 25-60% and for H bS variant concentration 25-60%. The diagnosis is sickle cell trait and not disease because of the low concentration in HbS. Furthermore, lane three has three bands that is also similar to the control sample, consisting HbS, HbF and HbA. As seen on lane three the bands are more merged then separated, making it difficult to distinguish the bands.
